circRNA-disease information
Expression pattern:
UP
Associated gene: KDM2A
Associated microRNA: miR-145-5p
Biological function: circGOLPH3 promotes OSCC cell growth, proliferation, migration, invasion, EMT, glycolysis and tumor growth, and inhibits apoptosis.
Molecular mechanism: circGOLPH3 acts as a cytoplasmic ceRNA sponge for miR-145-5p, thereby competitively inhibiting miR-145-5p-mediated repression of KDM2A and promoting OSCC malignant behaviors and glycolysis.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); EMT (promotes); glycolysis (promotes); apoptosis (inhibits); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Bioinformatics Analysis; Luciferase Reporter Assay; Transfection; Colony Formation Assay; EdU Staining; Transwell Assay; Western Blot; Annexin V/PI Flow Cytometry; In Vivo Animal Model; IHC (Immunohistochemistry)

Clinical significance:

circGOLPH3 was elevated in OSCC tissues and correlated with TNM staging; it may be a new target for OSCC pharmacologic therapy.

Description:

circGOLPH3 is upregulated in OSCC tissues and cell lines and is associated with higher tumor stage. Functionally, circGOLPH3 acts as a pro-oncogenic circRNA that promotes OSCC proliferation, migration, invasion, EMT, glycolysis and tumor growth while suppressing apoptosis. Mechanistically, cytoplasmic circGOLPH3 sponges miR-145-5p to increase KDM2A expression through a ceRNA mechanism.

Confidence score:

0.8174

Other information
Title:

Knockdown of circGOLPH3 inhibits cell progression and glycolysis by targeting miR-145-5p/lysine demethylase 2A (KDM2A) axis in oral squamous cell carcinoma.
