| Associated gene: |
c-Myc, CXCR4 |
| Associated microRNA: |

| Biological function: |
Promotes proliferation and migration and inhibits apoptosis in AML cells. |
| Molecular mechanism: |
Stabilizes c-Myc protein by reducing phosphorylation at Thr-58, thereby promoting downstream CXCR4 expression; c-Myc overexpression partially rescues circPVT1-knockdown phenotypes. |
| Biological pathway or process: |
proliferation (promotes); migration (promotes); apoptosis (inhibits); other pathway/process (promotes) |

| Validation methods: |
RT-qPCR; RNase R Treatment; Transfection; CCK8; Transwell Assay; Annexin V/PI Flow Cytometry; Western Blot; Clinical Sample Validation; Survival Analysis |
| Clinical significance: |
Higher expression of circPVT1 was associated with shorter overall survival (OS) and relapse-free survival (RFS) in AML patients. |
| Description: |
circPVT1 is up-regulated in AML bone marrow samples and AML cell lines and its high expression is associated with shorter OS and RFS. Functionally, circPVT1 promotes THP-1 cell proliferation and migration and inhibits apoptosis. Mechanistically, circPVT1 stabilizes c-Myc protein (via reduced Thr-58 phosphorylation) and enhances downstream CXCR4 expression, and c-Myc overexpression partially rescues circPVT1 knockdown effects. |
