| Associated gene: |
Ago2, DNMT3a, SOCS3, JAK2, STAT3, EP300, WDR5 |
| Associated microRNA: |
miR-502-5p |
| Biological function: |
promotes liver cancer cell growth/proliferation and migration; promotes cell cycle progression; inhibits apoptosis; promotes tumor growth in vivo |
| Molecular mechanism: |
EP300/WDR5-mediated histone modifications (H3K27ac, H3K4me3) activate circSOD2 transcription; circSOD2 sponges miR-502-5p to upregulate DNMT3a, which methylates SOCS3 promoter to suppress SOCS3, thereby activating JAK2/STAT3; STAT3 binds circSOD2 promoter creating a positive feedback loop |
| Biological pathway or process: |
JAK/STAT (promotes); proliferation (promotes); migration (promotes); cell cycle (promotes); apoptosis (inhibits); ceRNA regulation (promotes) |

RT-qPCR; divergent primers PCR; RNase R Treatment; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; ChIP / ChIP-seq; Western Blot; Transfection; CCK8; Cell Cycle Assay; Annexin V/PI Flow Cytometry; Transwell Assay; In Vivo Animal Model; Clinical Sample Validation; Survival Analysis; Bioinformatics Analysis |
| Clinical significance: |
High circSOD2 expression was associated with poor survival and linked to higher grade tumors |
| Description: |
circSOD2 (hsa_circ_0004662) is upregulated in HCC tissues/cell lines and promotes proliferation, migration, cell-cycle progression, and xenograft tumor growth while inhibiting apoptosis. Mechanistically, EP300/WDR5 activate circSOD2 via H3K27ac/H3K4me3 at its promoter, and circSOD2 sponges miR-502-5p to increase DNMT3a, which methylates and suppresses SOCS3 to activate JAK2/STAT3, with STAT3 feeding back to upregulate circSOD2. High circSOD2 expression is associated with poor patient survival. |
