| Associated gene: |
bone morphogenetic protein type 2 receptor, BMPR2 |
| Associated microRNA: |
miR-1256 |
| Biological function: |
Circ_0003506 promotes radioresistance, cell proliferation, migration and invasion, and inhibits apoptosis in radioresistant gastric cancer cells; knockdown of circ_0003506 enhances radiosensitivity in vivo. |
| Molecular mechanism: |
Circ_0003506 acts as a ceRNA sponge for miR-1256 and upregulates BMPR2 expression through the miR-1256/BMPR2 axis, thereby promoting gastric cancer radioresistance and malignant behaviors. |
| Biological pathway or process: |
radioresistance (promotes); proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); ceRNA regulation (other) |

RT-qPCR; Clinical Sample Validation; Nuclear-Cytoplasmic Fractionation; Luciferase Reporter Assay; Transfection; CCK8; Colony Formation Assay; Annexin V/PI Flow Cytometry; Transwell Assay; Western Blot; In Vivo Animal Model; Bioinformatics Analysis |
| Clinical significance: |
Circ_0003506 might become a diagnostic marker for radioresistance emergence and a therapeutic target for restoration of radiosensitivity in GC. |
| Description: |
This study found that hsa_circ_0003506/circ_0003506 is upregulated in gastric cancer, especially in radioresistant GC tissues and cells. Circ_0003506 promotes GC radioresistance and malignant phenotypes by sponging miR-1256 and increasing BMPR2 expression; its knockdown suppresses proliferation, migration and invasion, promotes apoptosis, and enhances radiosensitivity in vitro and in vivo. The authors propose circ_0003506 as a potential diagnostic marker for radioresistance emergence and therapeutic target for restoring radiosensitivity in GC. |
