circRNA-disease information
Expression pattern:
DN
Associated gene: ABCG1, Ago2
Associated microRNA: miR-208b-5p
Biological function: circ_0001445 alleviates ox-LDL-induced endothelial injury in HAECs by promoting cell viability, proliferation and migration, while repressing inflammatory cytokine release and extracellular matrix accumulation.
Molecular mechanism: circ_0001445 functions as a miR-208b-5p sponge and regulates ABCG1 expression through the circ_0001445/miR-208b-5p/ABCG1 ceRNA axis.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); migration (promotes); inflammation (inhibits); fibrosis (inhibits); other pathway/process (inhibits)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; Clinical Sample Validation; Transfection; ELISA; CCK8; EdU Staining; Transwell Assay; Western Blot; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; ROC Analysis; Bioinformatics Analysis

Clinical significance:

circ_0001445 was downregulated in CAD patient serum and showed diagnostic potential for CAD with an AUC of 0.9369; it might be a potential therapeutic target through the circ_0001445/miR-208b-5p/ABCG1 pathway.

Description:

circ_0001445 is a SMARCA5-derived human circRNA that is downregulated in CAD patient serum and ox-LDL-treated HAECs. Its overexpression protects HAECs from ox-LDL-induced endothelial injury by enhancing proliferation and migration and reducing inflammation and ECM accumulation, acting through the miR-208b-5p/ABCG1 axis. The study also suggests circ_0001445 has diagnostic potential for CAD.

Confidence score:

0.8166
