| Expression pattern: |
UP |
| Associated gene: |
KLF8 |
| Associated microRNA: |
miR-24-3p |
| Biological function: |
Promotes proliferation and chemoresistance of osteosarcoma cells; high expression correlates with larger tumor size, advanced TNM stage, and shorter overall survival. |
| Molecular mechanism: |
circPVT1 acts as a miR-24-3p sponge and up-regulates KLF8, thereby promoting proliferation and chemoresistance of osteosarcoma cells. |
| Biological pathway or process: |
proliferation (promotes); chemoresistance (promotes); drug resistance (promotes); ceRNA regulation (promotes) |

| Validation methods: |
RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Luciferase Reporter Assay; RNA Pull-Down; Western Blot; Survival Analysis; Bioinformatics Analysis |
| Clinical significance: |
High circPVT1 expression was correlated with tumor size, TNM stage, and shorter 5-year overall survival; circPVT1 overexpression might serve as an adverse prognostic factor in OS. |
| Description: |
circPVT1 is up-regulated in osteosarcoma tissues, osteosarcoma cell lines, and chemoresistant osteosarcoma cells. It promotes proliferation and resistance to cisplatin, doxorubicin, and methotrexate by sponging miR-24-3p and up-regulating KLF8. Clinically, high circPVT1 expression is associated with adverse clinicopathological features and poorer overall survival. |
