circRNA-disease information
Expression pattern:
UP
Associated gene: MET, c-MET, PI3K, AKT, Bax, Bcl2, E-cadherin, Vimentin, Ki67
Associated microRNA: miR-532-3p
Biological function: Promotes glioma cell proliferation, colony formation, invasion, EMT, metastasis and tumor growth, while inhibiting apoptosis.
Molecular mechanism: circFAM53B acts as a ceRNA that sponges miR-532-3p, thereby relieving miR-532-3p-mediated repression of MET/c-MET and activating the c-MET/PI3K/AKT pathway.
Biological pathway or process:

PI3K/AKT (promotes); proliferation (promotes); invasion (promotes); metastasis (promotes); EMT (promotes); apoptosis (inhibits); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Western Blot; Transwell Assay; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); In Vivo Animal Model; IHC (Immunohistochemistry); Survival Analysis; Bioinformatics Analysis

Clinical significance:

Higher circFAM53B levels were associated with poorer survival of glioma patients and the circFAM53B/miR-532-3p/c-MET/PI3K/AKT axis is proposed as a potential treatment target for glioma.

Description:

circFAM53B is up-regulated in glioma tissues and cell lines, and higher expression predicts poorer patient survival. Functionally, it promotes glioma proliferation, invasion, EMT and in vivo tumor growth while suppressing apoptosis. Mechanistically, circFAM53B sponges miR-532-3p to activate the MET/c-MET-mediated PI3K/AKT pathway.

Confidence score:

0.7789

Other information
Title:

CircFAM53B promotes the proliferation and metastasis of glioma through activating the c-MET/PI3K/AKT pathway via sponging miR-532-3p.
