circRNA-disease information
Expression pattern:
UP
Associated gene: FOXC2, METTL3
Associated microRNA: miR-199a
Biological function: circNRIP1 exerts oncogenic roles in osteosarcoma; circNRIP1 knockdown inhibits proliferation and migration and promotes apoptosis of osteosarcoma cells; high circNRIP1 expression is associated with TNM stage, distant metastasis, and shorter overall survival time.
Molecular mechanism: METTL3 elevates circNRIP1 expression via m6A modification; circNRIP1 functions as a miR-199a sponge and contributes to FOXC2 expression through a ceRNA mechanism.
Biological pathway or process:

proliferation (promotes); migration (promotes); apoptosis (inhibits); metastasis (promotes); ceRNA regulation (promotes); m6A modification (other)

Detected method:
Q
M
Validation methods:

Microarray; RNase R Treatment; RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Annexin V/PI Flow Cytometry; Transwell Assay; Luciferase Reporter Assay; MeRIP / MeRIP-seq; Bioinformatics Analysis; Survival Analysis; Cohort Study

Clinical significance:

High circNRIP1 expression is positively associated with TNM stage and distant metastasis and predicts shorter overall survival time in osteosarcoma patients.

Description:

circNRIP1 is up-regulated in osteosarcoma tissues and cell lines and acts as an oncogenic circRNA. Functionally, circNRIP1 promotes osteosarcoma cell proliferation and migration and suppresses apoptosis, while high expression is associated with advanced TNM stage, distant metastasis, and poor overall survival. Mechanistically, METTL3 increases circNRIP1 through m6A modification, and circNRIP1 sponges miR-199a to regulate FOXC2 expression.

Confidence score:

0.8323
