circRNA-disease information
Expression pattern:
UP
Associated gene: PIK3R3
Associated microRNA: miR-302a-3p
Biological function: circHN1 promotes CRC malignant behaviors, including proliferation, colony formation, invasion, migration and EMT, inhibits apoptosis, and counteracts curcumin-mediated suppression of CRC cells and tumor growth.
Molecular mechanism: circHN1 acts as a miR-302a-3p sponge to suppress miR-302a-3p and modulate PIK3R3, forming the circHN1/miR-302a-3p/PIK3R3 pathway involved in curcumin-mediated regulation of CRC malignancy.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); apoptosis (inhibits); migration (promotes); invasion (promotes); EMT (promotes); metastasis (promotes); drug resistance (promotes)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; Clinical Sample Validation; Luciferase Reporter Assay; RNA Pull-Down; Transfection; CCK8; Colony Formation Assay; Annexin V/PI Flow Cytometry; Transwell Assay; Western Blot; In Vivo Animal Model; Survival Analysis; Bioinformatics Analysis

Clinical significance:

Elevated circHN1 was associated with TNM staging, tumor size, lymph node metastasis and inferior overall survival rate; circHN1 was proposed as a promising CRC biomarker and potential therapeutic target.

Description:

circHN1 is up-regulated in human CRC tissues and cell lines and its high expression is associated with advanced clinical features and poor overall survival. Functionally, circHN1 promotes CRC malignant phenotypes and weakens curcumin-mediated antitumor effects by sponging miR-302a-3p and regulating PIK3R3.

Confidence score:

0.8005
