circRNA-disease information
Expression pattern:
UP
Associated gene: KLF12, Ago2, RISC
Associated microRNA: miR-15a-5p
Biological function: circ_0075503 promotes E2-induced viability, migration, invasion and epithelial-to-mesenchymal transition marker changes in eutopic endometrial stromal cells; knockdown suppresses these effects.
Molecular mechanism: circ_0075503 acts as a ceRNA by sponging miR-15a-5p to regulate KLF12 expression.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); EMT (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; Clinical Sample Validation; Transfection; MTT; Transwell Assay; Western Blot; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Bioinformatics Analysis

Clinical significance:

Knockdown of circ_0075503 indicates a novel target for the treatment of endometriosis.

Description:

circ_0075503 is up-regulated in ectopic endometrium and ectopic ESCs from endometriosis patients. Functionally, circ_0075503 promotes E2-induced ESC viability, migration, invasion and EMT-like marker changes, while its knockdown suppresses these phenotypes. Mechanistically, circ_0075503 sponges miR-15a-5p to increase KLF12 expression, forming a circ_0075503/miR-15a-5p/KLF12 regulatory axis with potential therapeutic relevance.
