circRNA-disease information
Expression pattern:
UP
Associated gene: STAT3
Associated microRNA: miR-365a-3p
Biological function: Promotes breast cancer cell proliferation/viability, migration/invasion, EMT and cancer stem cell properties; mediates propofol effects on tumor growth.
Molecular mechanism: Acts as a ceRNA sponge for miR-365a-3p to enhance STAT3 expression; STAT3 binds circNOLC1 promoter and propofol inhibits this STAT3-dependent transcription in a feedback loop.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); EMT (promotes); stemness (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Transwell Assay; Wound Healing Assay; Western Blot; Flow Cytometry(Non-apoptosis/cycle); Luciferase Reporter Assay; RNA Pull-Down; ChIP / ChIP-seq; In Vivo Animal Model; IHC (Immunohistochemistry)

Clinical significance:

Description:

circNOLC1 is up-regulated in breast cancer and promotes proliferation, migration/invasion, EMT, and cancer stem cell properties. Mechanistically, it acts as a ceRNA for miR-365a-3p to increase STAT3, and propofol suppresses circNOLC1 via inhibiting STAT3 recruitment to the circNOLC1 promoter, with circNOLC1 overexpression rescuing propofol’s anti-tumor effects in vitro and in vivo.

Confidence score:

0.7589

Other information
