circRNA-disease information
Expression pattern:
UP
Associated gene: TLR4, NLRP3, p-p65, p-IkappaBalpha, C-Caspase-1, IL-1beta, TNF-alpha
Associated microRNA: miR-192, miR-561
Biological function: Promotes inflammatory cytokine production and inflammatory response in gouty arthritis; circHIPK3 knockdown suppresses gouty arthritis in vivo.
Molecular mechanism: circHIPK3 acts as a miRNA sponge for miR-192 and miR-561, thereby promoting TLR4 and NLRP3 expression and activating inflammatory signaling pathways.
Biological pathway or process:

NF-kappaB (promotes); inflammation (promotes); immune regulation (promotes); ceRNA regulation (promotes); other pathway/process (promotes)

Detected method:
Q
S
Validation methods:

RT-qPCR; RNA-seq; Clinical Sample Validation; Transfection; ELISA; Western Blot; RNA Pull-Down; FISH / smFISH; Luciferase Reporter Assay; In Vivo Animal Model; Bioinformatics Analysis

Clinical significance:

circHIPK3 was elevated in SFMCs from patients with gouty arthritis, positively correlated with serum uric acid level, and may be a promising treatment target.

Description:

circHIPK3 is up-regulated in SFMCs from patients with gouty arthritis and in MSU-stimulated THP-1 cells. It promotes inflammatory cytokine production and inflammatory signaling by sponging miR-192 and miR-561, thereby increasing TLR4 and NLRP3 expression. Knockdown of circHIPK3 suppresses gouty arthritis in an MSU-induced mouse model, suggesting therapeutic potential.

Confidence score:

0.7372
