circRNA-disease information
Expression pattern:
UP
Associated gene: SNCA, AGO2, JAK2, STAT3
Associated microRNA: miR-7-5p
Biological function: circZNF208 promotes proliferation and X-ray radioresistance in NSCLC cells; knockdown of circZNF208 inhibits proliferation and enhances sensitivity to X-rays, but does not significantly alter sensitivity to carbon ions.
Molecular mechanism: circZNF208 acts as a competing endogenous RNA by sponging miR-7-5p, upregulates SNCA, and is linked to activation of the JAK2-STAT3 signalling pathway to regulate X-ray radiosensitivity in NSCLC cells.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); apoptosis (inhibits); JAK/STAT (promotes); radioresistance (promotes)

Detected method:
Q
S
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; RNA-seq; Nuclear-Cytoplasmic Fractionation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; Annexin V/PI Flow Cytometry; Western Blot; Bioinformatics Analysis

Clinical significance:

circZNF208 might serve as a potential biomarker and therapeutic target for selecting radiotherapy modalities in NSCLC.

Description:

In NSCLC radioresistant A549-R11 cells, circZNF208 is upregulated and promotes resistance to low-LET X-ray irradiation. It functions as a ceRNA sponge for miR-7-5p, leading to increased SNCA expression and activation of the JAK2-STAT3-related pathway. circZNF208 knockdown sensitizes NSCLC cells to X-rays but not to carbon-ion irradiation, suggesting potential value as a biomarker for radiotherapy modality selection.

Confidence score:

0.8161

Other information
Title:

CircZNF208 enhances the sensitivity to X-rays instead of carbon-ions through the miR-7-5p /SNCA signal axis in non-small-cell lung cancer cells.
