| Expression pattern: |
DN |
| Associated gene: |
PTEN, DHX9, Alu repeat elements, PI3K/AKT signaling pathway |
| Associated microRNA: |
miR-155, miR-330-3p |
| Biological function: |
Circ-PTEN functions as a tumor-inhibiting circRNA in NSCLC, inhibiting NSCLC cell proliferation in vitro and retarding tumor growth in vivo. |
| Molecular mechanism: |
DHX9 upregulation reduces circ-PTEN production; circ-PTEN acts as a miRNA sponge for miR-155 and miR-330-3p to elevate PTEN and inactivate PI3K/AKT signaling. |
| Biological pathway or process: |
PI3K/AKT (inhibits); proliferation (inhibits); ceRNA regulation (promotes); mRNA stability (other); other pathway/process (inhibits) |

| Validation methods: |
RNase R Treatment; RT-qPCR; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; In Vivo Animal Model; IHC (Immunohistochemistry); Western Blot; Survival Analysis; ROC Analysis; Bioinformatics Analysis; Nuclear-Cytoplasmic Fractionation |
| Clinical significance: |
Low circ-PTEN was linked with malignant clinical features and poor outcome; high circ-PTEN was positively correlated with small tumor size, early TNM stage and favorable prognosis; serum circ-PTEN showed diagnostic value for NSCLC with AUC 0.8356. |
| Description: |
Circ-PTEN (hsa_circ_0094342), a PTEN-derived circRNA, is down-regulated in NSCLC tissues and serum and its low expression is associated with malignant clinicopathological features and poor outcome. Functionally, circ-PTEN suppresses NSCLC cell proliferation and xenograft tumor growth. Mechanistically, DHX9 reduces circ-PTEN production, while circ-PTEN sponges miR-155 and miR-330-3p to increase PTEN expression and inhibit PI3K/AKT signaling, supporting its potential diagnostic, prognostic and therapeutic value. |
