circRNA-disease information
Expression pattern:
UP
Associated gene: CLIC4, ARHGAP12
Associated microRNA: miR-135a-5p
Biological function: Circ_0000231 contributed to ox-LDL-induced injury of HUVECs, antagonized the protective role of ASV, reduced cell viability and migration, and promoted apoptosis, inflammatory response, oxidative stress, MCP1 and ICAM1 expression.
Molecular mechanism: Circ_0000231 acts as a miRNA sponge for miR-135a-5p and regulates CLIC4 expression through the circ_0000231/miR-135a-5p/CLIC4 axis in ox-LDL-induced HUVECs.
Biological pathway or process:

ceRNA regulation (other); apoptosis (promotes); migration (inhibits); inflammation (promotes); other pathway/process (promotes)

Detected method:
Q
Validation methods:

Actinomycin D / DRB Stability Assay; RNase R Treatment; RT-qPCR; Nuclear-Cytoplasmic Fractionation; Transfection; MTT; Annexin V/PI Flow Cytometry; Transwell Assay; Wound Healing Assay; ELISA; Western Blot; Luciferase Reporter Assay; RNA Pull-Down; Bioinformatics Analysis

Clinical significance:

Description:

In an ox-LDL-induced HUVEC atherosclerosis cell model, circ_0000231 was up-regulated and promoted endothelial cell injury, including apoptosis, inflammation, oxidative stress and impaired migration/viability. Mechanistically, circ_0000231 sponged miR-135a-5p to regulate CLIC4, and ASV alleviated cell injury by targeting the circ_0000231/miR-135a-5p/CLIC4 axis.

Confidence score:

0.763
