circRNA-disease information
Expression pattern:
DN
Associated gene: Cyclin F, MYBL2, PLK1, CCND1, CDK6, Cyclin A
Associated microRNA: -
Biological function: circ-MYBL2 functions as a tumor suppressor in MM by repressing MM cell viability, DNA synthesis, cell cycle progression, proliferation, and in vivo tumor growth.
Molecular mechanism: circ-MYBL2 directly binds Cyclin F and MYBL2 protein, facilitates Cyclin F-MYBL2 interaction, decreases MYBL2 phosphorylation and activation, and inhibits transcription of proliferation-related MYBL2 target genes.
Biological pathway or process:

proliferation (inhibits); cell cycle (inhibits); other pathway/process (other)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Clinical Sample Validation; Transfection; CCK8; EdU Staining; Cell Cycle Assay; RNA Pull-Down; RIP (RNA Immunoprecipitation); Western Blot; Co-IP; In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); Cohort Study; Survival Analysis; ROC Analysis; Bioinformatics Analysis

Clinical significance:

Low circ-MYBL2 level was associated with high clinical stage and unfavorable outcome; high circ-MYBL2 expression predicted longer overall survival; serum circ-MYBL2 showed high diagnostic accuracy for MM with AUC 0.8394.

Description:

circ-MYBL2 is down-regulated in MM tissues and serum and acts as a tumor suppressor. It inhibits MM cell proliferation and cell cycle progression by binding Cyclin F and enhancing Cyclin F interaction with MYBL2, leading to reduced MYBL2 phosphorylation and decreased transcription of proliferation-related targets. Clinically, low circ-MYBL2 is associated with advanced stage and poorer survival, while serum circ-MYBL2 shows diagnostic potential.

Confidence score:

0.8679
