circRNA-disease information
Expression pattern:
DN
Associated gene: PRKCD / protein kinase C, delta
Associated microRNA: miR-615-3p
Biological function: circITCH inhibits MM cell proliferation, promotes apoptosis of BTZ-resistant MM cells, and enhances sensitivity to bortezomib.
Molecular mechanism: circITCH functions as a miRNA sponge for miR-615-3p and regulates PRKCD expression, thereby enhancing bortezomib sensitivity in MM cells.
Biological pathway or process:

proliferation (inhibits); apoptosis (promotes); chemoresistance (inhibits); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Survival Analysis; Bioinformatics Analysis; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Transfection; MTT; Annexin V/PI Flow Cytometry; Western Blot; In Vivo Animal Model

Clinical significance:

Reduced expression of circITCH is indicative of poor prognosis in MM patients; high circITCH expression is associated with longer overall survival.

Description:

circITCH is downregulated in multiple myeloma bone marrow specimens, MM cell lines, and bortezomib-resistant MM cells. Higher circITCH expression predicts longer overall survival, and circITCH overexpression enhances bortezomib sensitivity by promoting apoptosis and suppressing viability through the miR-615-3p/PRKCD axis.

Confidence score:

0.7537
