circRNA-disease information
Expression pattern:
UP
Associated gene: SOX4
Associated microRNA: miRNA-338-3p
Biological function: Promotes NSCLC cell proliferation, invasion/metastasis, and drug resistance; circ_0005909 knockdown suppresses these behaviors and reduces xenograft tumor growth.
Molecular mechanism: Acts as a ceRNA/miRNA sponge for miRNA-338-3p, thereby relieving repression of SOX4.
Biological pathway or process:

proliferation (promotes); invasion (promotes); metastasis (promotes); drug resistance (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; EdU Staining; Transwell Assay; In Vivo Animal Model; Luciferase Reporter Assay; RNA Pull-Down; Western Blot; Cohort Study; Survival Analysis; Bioinformatics Analysis

Clinical significance:

High circ_0005909 expression is associated with poor prognosis and shorter overall survival in NSCLC patients; independently associated with overall survival.

Description:

hsa_circ_0005909 (circ_0005909), derived from XPR1, is up-regulated in NSCLC tissues and cell lines and its high expression predicts shorter overall survival. Functionally it promotes proliferation, invasion/metastasis and adriamycin-related drug resistance, and its knockdown inhibits xenograft tumor growth. Mechanistically, it acts as a ceRNA by sponging miRNA-338-3p to increase SOX4 expression.

Confidence score:

0.8353
