circRNA-disease information
Expression pattern:
DN
Associated gene: MBD2, FGFR4, NFYA
Associated microRNA: miR-520c-3p
Biological function: Suppresses HCC recurrence and inhibits HCC cell proliferation, colony formation, migration, and tumor formation; modulates lenvatinib response.
Molecular mechanism: Transcriptionally repressed by NFYA; acts as a miRNA sponge for miR-520c-3p to regulate MBD2 and downstream FGFR4.
Biological pathway or process:

proliferation (inhibits); migration (inhibits); cell cycle (inhibits); apoptosis (promotes); ceRNA regulation (other); other pathway/process (inhibits); drug resistance (other)

Detected method:
Q
S
Validation methods:

circRNA-seq; RNA-seq; Bioinformatics Analysis; Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RT-qPCR; Clinical Sample Validation; Cohort Study; Survival Analysis; circRIP; RNA Pull-Down; Luciferase Reporter Assay; ChIP / ChIP-seq; Transfection; CCK8; Colony Formation Assay; Transwell Assay; Cell Cycle Assay; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model; H&E Staining; Annexin V/PI Flow Cytometry

Clinical significance:

Lower circKCNN2 in tumors is associated with recurrence; elevated circKCNN2 in tumors predicts favorable overall survival and recurrence-free survival and is associated with decreased risk of HCC recurrence.

Description:

circKCNN2 is downregulated in HCC and acts as a tumor-suppressive circRNA associated with lower recurrence risk and better survival. Mechanistically, it is repressed by NFYA and suppresses HCC progression by sponging miR-520c-3p to modulate MBD2 and downstream FGFR4, influencing lenvatinib response.

Confidence score:

0.8913

Other information
Title:

circKCNN2 suppresses the recurrence of hepatocellular carcinoma at least partially via regulating miR-520c-3p/methyl-DNA-binding domain protein 2 axis.
