circRNA-disease information
Expression pattern:
UP
Associated gene: USP28, AGO2
Associated microRNA: miR-655-3p
Biological function: promotes lenvatinib resistance in HCC; promotes tumor growth under lenvatinib treatment
Molecular mechanism: ceRNA mechanism: circMED27 sponges miR-655-3p to upregulate USP28; association with AGO2 complex
Biological pathway or process:

ceRNA regulation (promotes); drug resistance (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Survival Analysis; Cohort Study; FISH / smFISH; Luciferase Reporter Assay; circRIP; RIP (RNA Immunoprecipitation); Transfection; CCK8; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model

Clinical significance:

Higher levels of circMED27 were correlated with bad clinical characteristics and poor prognoses of patients with HCC; serum exosomal circMED27 is an independent predictor for OS and postoperative recurrence; high circMED27 predicts shorter PFS under lenvatinib

Description:

circMED27 is upregulated in HCC tissues and serum exosomes and is associated with worse prognosis. Functionally, circMED27 promotes lenvatinib resistance by acting as a ceRNA that sponges miR-655-3p, thereby increasing USP28 expression, with predominant cytoplasmic localization and validation in HCC cells and xenografts.

Confidence score:

0.8062
