circRNA-disease information
Expression pattern:
UP
Associated gene: LDHA, FGFR1, AGO2
Associated microRNA: miR-511-5p
Biological function: promotes CRC cell proliferation, inhibits apoptosis, promotes tumor growth in vivo, promotes the Warburg effect and glycolytic metabolism, and reduces OXPHOS activity
Molecular mechanism: circTATDN3 acts as an adaptor/scaffold molecule enhancing LDHA-FGFR1 binding and FGFR1-induced LDHA phosphorylation; it also sponges miR-511-5p to increase LDHA expression, synergistically promoting CRC progression and the Warburg effect
Biological pathway or process:

glycolysis (promotes); proliferation (promotes); apoptosis (inhibits); oxidative phosphorylation (inhibits); ceRNA regulation (promotes); other pathway/process (promotes)

Detected method:
Q
M
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Actinomycin D / DRB Stability Assay; RT-qPCR; Microarray; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Co-IP; IF (Immunofluorescence); Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; Annexin V/PI Flow Cytometry; IHC (Immunohistochemistry); In Vivo Animal Model; Western Blot; Survival Analysis; Cohort Study; Bioinformatics Analysis

Clinical significance:

high circTATDN3 expression was positively associated with poor postoperative prognosis and was an independent prognostic factor for CRC

Description:

circTATDN3 is up-regulated in colorectal cancer cells and tumor tissues, and high expression is associated with poor postoperative prognosis. It promotes CRC proliferation, tumor growth, and the Warburg effect by scaffolding LDHA and FGFR1 to enhance LDHA phosphorylation and by sponging miR-511-5p to increase LDHA expression. These findings suggest circTATDN3 as a potential therapeutic target and prognostic biomarker in CRC.
