| Biological function: |
circPVT1 is upregulated in CRC and functions as an oncogenic circRNA; its encoded product PVT1-104aa promotes CRC proliferation, colony formation, migration, metastasis, tumor growth, immune evasion, and response to combined anti-PD-L1 therapy. |
| Molecular mechanism: |
circPVT1 encodes PVT1-104aa via IRES-mediated translation; PVT1-104aa binds c-Myc, disrupts c-Myc interaction with GSK3beta and FBW7, suppresses ubiquitination-mediated c-Myc degradation, and increases PD-L1 expression through c-Myc-dependent transcriptional regulation. |

| Clinical significance: |
PVT1-104aa upregulation is linked to deteriorated prognosis among individuals with CRC; elevated PVT1-104aa levels correlated significantly with adverse clinical outcomes, indicating its potential as a prognostic biomarker and therapeutic target. |
| Description: |
circPVT1 is a PVT1-derived circRNA that is upregulated in colorectal cancer tissues and cell lines. Its oncogenic effect is mediated mainly through translation of PVT1-104aa, which stabilizes c-Myc by inhibiting GSK3beta/FBW7-associated ubiquitination and thereby increases PD-L1 expression, promoting tumor growth, metastasis, immune evasion, and poorer prognosis. PVT1-104aa inhibition enhances anti-PD-L1 treatment efficacy in syngeneic CRC models. |
