circRNA-disease information
Expression pattern:
UP
Associated gene: IL6, FOS, ITGB3, BCL2L11, MAP2K1, HDAC2, PDGFRB, HNRNPA1, CARM1, BPTF, CD44, SOX2, OCT4
Associated microRNA: miR-149
Biological function: circTCF25 promotes gastric cancer cell proliferation, migration, invasion and cancer stem cell marker expression, and its knockdown represses cancer cell viability, migration, invasion and stemness-associated markers.
Molecular mechanism: circTCF25 appears to act as a miR-149 sponge and may regulate malignant activities in gastric cancer by targeting miR-149 and subsequently modulating downstream mRNA expression, including genes enriched in PI3K-AKT, MAPK and EGFR tyrosine kinase inhibitor resistance-related pathways.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); stemness (promotes); ceRNA regulation (promotes); PI3K/AKT (other); MAPK (other); EGFR (other); drug resistance (other)

Detected method:

Validation methods:

RT-qPCR; Clinical Sample Validation; Cohort Study; Survival Analysis; Transfection; CCK8; Transwell Assay; Luciferase Reporter Assay; Bioinformatics Analysis

Clinical significance:

circTCF25 was associated with lymph node metastasis, T stage, TNM stage and shorter overall survival; increased expression may serve as a biomarker to predict survival outcome in patients with gastric cancer.

Description:

In this study, circTCF25/hsa_circ_0041103 was up-regulated in human gastric cancer tissues and cell lines and high expression predicted shorter overall survival. Functional knockdown experiments indicated that circTCF25 promotes gastric cancer cell proliferation, migration, invasion and CSC marker expression. Mechanistically, the study supports a circTCF25-miR-149 regulatory network with predicted downstream mRNAs and cancer-related pathways such as PI3K-AKT and MAPK.
