circRNA-disease information
Expression pattern:
UP
Associated gene: CBX8
Associated microRNA: miR-1236-3p
Biological function: circSPECC1 promotes BC cell growth, proliferation and long-term survival and suppresses apoptosis; circSPECC1 knockdown suppresses tumor growth in vitro and in vivo and promotes apoptosis under irradiated and nonirradiated conditions.
Molecular mechanism: circSPECC1 functions as a ceRNA for miR-1236-3p to regulate CBX8 expression, thereby affecting growth and apoptosis of BC cells.
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Transfection; MTT; EdU Staining; Colony Formation Assay; Annexin V/PI Flow Cytometry; Western Blot; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); In Vivo Animal Model; IHC (Immunohistochemistry); Clinical Sample Validation; Bioinformatics Analysis

Clinical significance:

Higher circSPECC1 expression was associated with tumor size, lymph node metastasis and TNM stage in BC patients.

Description:

This study shows that human circSPECC1 (hsa_circ_0000745), derived from SPECC1, is up-regulated in breast cancer tumors and cells and is further induced by radiation. Functionally, circSPECC1 supports BC cell growth and survival while suppressing apoptosis, and its knockdown inhibits xenograft tumor growth. Mechanistically, circSPECC1 acts through a miR-1236-3p/CBX8 regulatory axis and may represent a therapeutic target in BC.

Confidence score:

0.8298
