| Biological function: |
Regulates VEGFA mRNA splicing toward a reduced pro-angiogenic (Iso8a) to anti-angiogenic (Iso8b) isoform ratio and is associated with lower angiogenesis/microvessel density in GBM. |
| Molecular mechanism: |
circSMARCA5 binds the splicing factor SRSF1 (RBP sponge/tethering), thereby influencing alternative splicing of VEGFA pre-mRNA and the pro-/anti-angiogenic isoform switch. |

| Validation methods: |
RIP (RNA Immunoprecipitation); RT-qPCR; divergent primers PCR; Clinical Sample Validation; Transfection; Survival Analysis; Bioinformatics Analysis |
| Clinical significance: |
Low circSMARCA5 expression is associated with poorer overall survival and progression-free survival in GBM; circSMARCA5 is proposed as a prognostic biomarker and prospective therapeutic target. |
| Description: |
circSMARCA5 is down-regulated in GBM and physically binds the splicing factor SRSF1. By tethering SRSF1, circSMARCA5 modulates VEGFA alternative splicing, reducing the pro-angiogenic/anti-angiogenic VEGFA isoform ratio and associating with lower microvessel density. Higher circSMARCA5 expression indicates better OS/PFS, supporting its prognostic value and potential anti-angiogenic therapeutic relevance in GBM. |
