Validation methods:

RNA-seq; Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RT-qPCR; Clinical Sample Validation; Transfection; MTT; Survival Analysis

Clinical significance:

Circular, and not linear, CNOT2 levels are predictive for progression-free survival time to aromatase inhibitor (AI) therapy in advanced breast cancer patients; circCNOT2 is detectable in cell-free RNA from plasma.

Description:

circCNOT2 is present in primary breast tumors and detectable in plasma cfRNA. Functional knockdown of circCNOT2 reduces viability of MCF-7 and BT-474 breast cancer cells. In ER-positive advanced breast cancer treated with aromatase inhibitors, higher circCNOT2 levels predict shorter progression-free survival, while linear CNOT2 does not.

0.7286
