circRNA-disease information
Expression pattern:
UP
Associated gene: ITGA6, PI3K, AKT, Ago2
Associated microRNA: miR-30e-5p
Biological function: Promotes NSCLC cell proliferation, migration, invasion and EMT, inhibits apoptosis, and accelerates tumor growth in vivo.
Molecular mechanism: Acts as a ceRNA/miRNA sponge for miR-30e-5p to upregulate ITGA6 and activate the ITGA6/PI3K/AKT axis.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); EMT (promotes); apoptosis (inhibits); PI3K/AKT (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Transfection; CCK8; Colony Formation Assay; TUNEL; Transwell Assay; Western Blot; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; In Vivo Animal Model; IHC (Immunohistochemistry); Survival Analysis; Bioinformatics Analysis

Clinical significance:

circ-PITX1 up-regulation was correlated with NSCLC patients’ poor survival; high circ-PITX1 associated with higher TNM stage and earlier local lymph node metastasis.

Description:

circ-PITX1 is up-regulated in NSCLC and promotes malignant phenotypes (proliferation, migration/invasion, EMT) while inhibiting apoptosis. It functions mainly as a ceRNA that sponges miR-30e-5p to relieve repression of ITGA6, thereby activating the ITGA6/PI3K/AKT pathway and correlating with poor patient survival.

Confidence score:

0.798
