| Associated gene: |
Bim / Bcl-2-interacting mediator of cell death |
| Associated microRNA: |
miR-423-3p |
| Biological function: |
Inhibits cell proliferation, viability, migration, EMT and tumorigenesis; induces apoptosis in hepatoblastoma. |
| Molecular mechanism: |
Acts as a ceRNA sponge for miR-423-3p to positively regulate Bim. |
| Biological pathway or process: |
proliferation (inhibits); migration (inhibits); EMT (inhibits); apoptosis (promotes); ceRNA regulation (other); metastasis (inhibits) |

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Annexin V/PI Flow Cytometry; Transwell Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Western Blot; In Vivo Animal Model; Survival Analysis |
| Clinical significance: |
Low expression of circSETD3 associated with poor survival of HB patients; circSETD3 downregulated in advanced clinical stage compared to early clinical stage. |
| Description: |
circSETD3 is down-regulated in hepatoblastoma and acts as a tumor suppressor. It mainly localizes to the cytoplasm and inhibits HB cell proliferation/migration/EMT while promoting apoptosis by sponging miR-423-3p to upregulate the pro-apoptotic target Bim, suppressing tumor growth in a cell line-derived xenograft model. |
