Validation methods:

Microarray; RNase R Treatment; RT-qPCR; FISH / smFISH; Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Colony Formation Assay; Transwell Assay; Wound Healing Assay; Annexin V/PI Flow Cytometry; Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry); TUNEL; Bioinformatics Analysis; Clinical Sample Validation

Clinical significance:

circ_PPAPDC1A may serve as a novel diagnostic biomarker and therapeutic target for NSCLC patients with Osimertinib resistance.

Description:

circ_PPAPDC1A is up-regulated in Osimertinib-acquired resistant NSCLC tissues/cells and promotes resistance by enhancing proliferation, invasion and migration while inhibiting apoptosis. Mechanistically, it acts as a ceRNA sponging miR-30a-3p to upregulate IGF1R and activate the PI3K/AKT/mTOR pathway, and targeting circ_PPAPDC1A can partially reverse resistance.

Confidence score:

0.7956
