circRNA-disease information
Expression pattern:
UP
Associated gene: Ku70, DHX9, NOP14 mRNA
Associated microRNA: -
Biological function: Increases radiosensitivity of HCC cells to 125 I seed irradiation and external radiation; inhibits proliferation and migration; promotes apoptosis and irradiation-induced DNA damage.
Molecular mechanism: circNOP14 acts as a protein scaffold that interacts with Ku70 and prevents its nuclear translocation, thereby inhibiting Ku70-dependent DNA damage repair and increasing irradiation-induced DNA damage.
Biological pathway or process:

apoptosis (promotes); proliferation (inhibits); migration (inhibits); radioresistance (inhibits); other pathway/process (inhibits)

Detected method:

S
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; RT-qPCR; RNA-seq; FISH / smFISH; ISH (In Situ Hybridization); IHC (Immunohistochemistry); IF (Immunofluorescence); Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Transfection; CCK8; EdU Staining; Annexin V/PI Flow Cytometry; Transwell Assay; In Vivo Animal Model; Western Blot

Clinical significance:

High circNOP14 expression was associated with better objective response rate and short-term efficacy after 125 I seed implantation therapy; circNOP14 may serve as a predictive indicator, prognostic biomarker, radiosensitive target, and potential drug target for HCC radiotherapy.

Description:

circNOP14 is a stable cytoplasmic circRNA derived from NOP14 and is upregulated in HCC after 125 I seed irradiation. It enhances radiosensitivity to both 125 I brachytherapy and external radiation by binding Ku70, blocking Ku70 nuclear import, inhibiting DNA damage repair, and increasing radiation-induced DNA damage and apoptosis. Clinically, higher circNOP14 expression is associated with better objective response and short-term efficacy in HCC patients receiving 125 I seed implantation therapy.
