circRNA-disease information
Expression pattern:
DN
Associated gene: H2AX, gammaH2AX, p53, Bclaf1, Bcl2
Associated microRNA: -
Biological function: Inhibits proliferation and survival and promotes apoptosis in p53 mutant breast cancer cells; represses tumor growth and increases mouse survival.
Molecular mechanism: Forms protein complexes: in p53 wild-type cells circ-Ccnb1 binds H2AX and p53; in p53 mutant cells circ-Ccnb1 forms a complex with H2AX and Bclaf1 (not mutant p53), inducing cell death.
Biological pathway or process:

proliferation (inhibits); apoptosis (promotes); other pathway/process (other)

Validation methods:

Microarray; RNase R Treatment; RT-qPCR; Northern Blot; Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; Clinical Sample Validation; Nuclear-Cytoplasmic Fractionation; RNA Pull-Down; RIP (RNA Immunoprecipitation); CLIP; Western Blot; Transfection; MTT; Annexin V/PI Flow Cytometry; Colony Formation Assay; In Vivo Animal Model; Survival Analysis; ISH (In Situ Hybridization)

Clinical significance:

circ-Ccnb1 levels were decreased in breast carcinoma tissues and were significantly lower in p53 mutation tumors compared to tumors of wild-type p53.

Description:

circ-Ccnb1 is down-regulated in breast carcinoma tissues. Functionally, circ-Ccnb1 suppresses growth and promotes apoptosis of p53-mutant breast cancer cells by forming a circ-Ccnb1-H2AX-Bclaf1 complex, thereby inhibiting xenograft tumor growth and prolonging mouse survival.
