circRNA-disease information
Expression pattern:
UN
Associated gene: PES1, NOP14, BOP1, BRIX1, NOLC1, AGO2
Associated microRNA: miR-4659a/b, miR-3184-3p, miR-5571-5p
Biological function: confers atheroprotection by inducing apoptosis and inhibiting proliferation in vascular smooth muscle cells and macrophages
Molecular mechanism: circANRIL binds PES1, impairs exonuclease-mediated pre-rRNA processing, inhibits ribosome biogenesis, induces nucleolar stress and activates p53
Biological pathway or process:

p53 signaling (promotes); apoptosis (promotes); proliferation (inhibits); mitochondrial function (other); oxidative phosphorylation (other); mRNA stability (other); ribosome biogenesis (inhibits); other pathway/process (other)

Detected method:
Q
M
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; Sanger Sequencing; RT-qPCR; Microarray; RNase R Treatment; Actinomycin D / DRB Stability Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; Western Blot; Northern Blot; IF (Immunofluorescence); ISH (In Situ Hybridization); IHC (Immunohistochemistry); Clinical Sample Validation; Cohort Study; Bioinformatics Analysis; CCK8

Clinical significance:

High circANRIL expression is associated with less coronary artery disease (CAD) and CAD-protective 9p21 genotype.

Description:

circANRIL (from the ANRIL/CDKN2B-AS1 locus at 9p21) is associated with CAD-protective genotype and lower CAD burden. Mechanistically, it binds PES1 in the PeBoW complex, inhibits pre-rRNA processing and ribosome biogenesis, triggers nucleolar stress and p53 activation, thereby promoting apoptosis and suppressing proliferation in atherogenesis-relevant human vascular cells.

Confidence score:

0.8913
