circRNA-disease information
Expression pattern:
UP
Associated gene: RSF1, Ago2
Associated microRNA: miR-503-5p
Biological function: Promotes proliferation, tumor growth and cisplatin/DDP resistance; represses apoptosis; exosomal transfer confers DDP resistance to sensitive cervical cancer cells.
Molecular mechanism: Exosome-mediated transfer of circSYT15 enhances DDP resistance by acting as a miR-503-5p sponge to upregulate RSF1 expression.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); apoptosis (inhibits); chemoresistance (promotes); drug resistance (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Transfection; CCK8; EdU Staining; Colony Formation Assay; Annexin V/PI Flow Cytometry; RNA Pull-Down; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Western Blot; In Vivo Animal Model; IHC (Immunohistochemistry); Bioinformatics Analysis

Clinical significance:

Exosomal circSYT15 is correlated with DDP resistance in CC patients and is elevated in serum exosomes from DDP-resistant patients compared with DDP-sensitive patients.

Description:

circSYT15 is upregulated in DDP-resistant cervical cancer cells and serum/cell-derived exosomes. Exosome-delivered circSYT15 promotes proliferation, tumor growth and cisplatin resistance while inhibiting apoptosis, partly through sponging miR-503-5p and increasing RSF1 expression. Its elevated serum exosomal expression is associated with DDP resistance in cervical cancer patients.

Confidence score:

0.7754
