Associated gene: EIF4A3, ATG12
Associated microRNA: -
Biological function: Promotes K562/R cell viability, invasion, autophagy, tumor growth and Imatinib resistance, and inhibits apoptosis.
Molecular mechanism: circ_SIRT1 binds EIF4A3 and regulates EIF4A3-mediated ATG12 expression, thereby promoting autophagy and Imatinib resistance in CML.
Biological pathway or process:

proliferation (promotes); invasion (promotes); apoptosis (inhibits); autophagy (promotes); drug resistance (promotes); mRNA stability (promotes); other pathway/process (promotes)

Detected method:

Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; RT-qPCR; FISH / smFISH; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Actinomycin D / DRB Stability Assay; Transfection; CCK8; Annexin V/PI Flow Cytometry; Transwell Assay; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model; Bioinformatics Analysis

Clinical significance:

High expression of circ_SIRT1 is correlated with Imatinib resistance in CML and circ_SIRT1 may serve as a novel therapeutic target for Imatinib-resistant CML treatment.

Description:

circ_SIRT1 is a SIRT1-derived human circRNA that is up-regulated in CML, especially in Imatinib-resistant CML samples and K562/R cells. It promotes CML cell viability, invasion, autophagy, xenograft tumor growth and Imatinib resistance while suppressing apoptosis. Mechanistically, circ_SIRT1 binds EIF4A3 and increases ATG12 expression, linking the circ_SIRT1-EIF4A3-ATG12 mechanism to autophagy-associated Imatinib resistance.
