| Expression pattern: |

| Associated gene: |
IGF2BP1, CTNNB1 |
| Associated microRNA: |

| Biological function: |
promotes proliferation and migration/invasion; inhibits apoptosis; promotes tumor growth in vivo |
| Molecular mechanism: |
circXPO1 binds IGF2BP1 and enhances CTNNB1 mRNA stability, increasing CTNNB1 expression and promoting LUAD progression; circXPO1 overexpression is positively correlated with XPO1 gene copy number gain. |
| Biological pathway or process: |
proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); Wnt/beta-catenin (promotes); mRNA stability (promotes) |

RNA-seq; RT-qPCR; Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; Nuclear-Cytoplasmic Fractionation; RNA Pull-Down; RIP (RNA Immunoprecipitation); Actinomycin D / DRB Stability Assay; Transfection; MTT; EdU Staining; Colony Formation Assay; Transwell Assay; Wound Healing Assay; TUNEL; Western Blot; IHC (Immunohistochemistry); In Vivo Animal Model; Clinical Sample Validation; Cohort Study; Survival Analysis |
| Clinical significance: |
circXPO1 was overexpressed in LUAD tumor tissues and high circXPO1 expression was associated with shorter overall survival times; suggested as a biomarker for poor survival and a therapeutic target. |
| Description: |
circXPO1 (hsa_circ_0001016), derived from XPO1 exon 3-4, is upregulated in LUAD and correlates with worse overall survival. It predominantly localizes in the cytoplasm, binds the RBP IGF2BP1, enhances CTNNB1 mRNA stability and CTNNB1 protein expression, thereby promoting LUAD cell proliferation and migration/invasion and tumor growth; circXPO1 knockdown suppresses growth in xenograft and PDX models. |
