circRNA-disease information
Expression pattern:
UP
Associated gene: ERBB4, Ago2
Associated microRNA: miR-326
Biological function: Promotes NSCLC cell proliferation, viability, colony formation, migration, invasion, metastatic/aggressive behavior, and tumor growth; knockdown suppresses these malignant phenotypes.
Molecular mechanism: circRPPH1 functions as a ceRNA by sponging miR-326, thereby regulating ERBB4 expression through the miR-326/ERBB4 signal axis.
Biological pathway or process:

ceRNA regulation (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); EGFR (promotes)

Detected method:
Q
M
Validation methods:

Sanger Sequencing; RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Microarray; FISH / smFISH; Clinical Sample Validation; RIP (RNA Immunoprecipitation); RNA Pull-Down; Luciferase Reporter Assay; Transfection; CCK8; Colony Formation Assay; Transwell Assay; In Vivo Animal Model; Cohort Study; Survival Analysis; ROC Analysis; Bioinformatics Analysis

Clinical significance:

High circRPPH1 expression is associated with TNM stage, lymphatic metastasis, lower overall survival, and may serve as a diagnostic and prognostic biomarker for NSCLC.

Description:

circRPPH1 is up-regulated in NSCLC tissues and cells and is associated with advanced TNM stage, lymphatic metastasis, and poor overall survival. Functionally, circRPPH1 promotes NSCLC cell proliferation, migration, invasion, and xenograft tumor growth. Mechanistically, it acts through a ceRNA mechanism by sponging miR-326 and regulating ERBB4 expression via the miR-326/ERBB4 axis.

Confidence score:

0.8914
