JAK2, STAT3, p-STAT3, FUS, CTCF |
| Associated microRNA: |
miR-485-3p |
| Biological function: |
promotes proliferation, migration and invasion; enhances tumour growth and metastasis |
| Molecular mechanism: |
Acts as a ceRNA to sponge miR-485-3p and upregulate JAK2, increasing p-STAT3; directly interacts with and recruits FUS to facilitate nuclear translocation of p-STAT3; transcriptionally enhanced by CTCF |

High expression of circSPARC was associated with advanced TNM stage, lymph node metastases, and poor survival; circSPARC might serve as a potential diagnostic and prognostic biomarker |
| Description: |
circSPARC (hsa_circ_0004104), derived from SPARC, is upregulated in CRC tissues and plasma and promotes CRC proliferation, migration, invasion, tumor growth and metastasis. It acts as a cytoplasmic ceRNA sponging miR-485-3p to increase JAK2 expression and enhance JAK2/STAT3 signaling (increased p-STAT3), and also recruits the RBP FUS to facilitate nuclear translocation of p-STAT3. High circSPARC is associated with advanced clinicopathologic features and poorer overall survival, with potential diagnostic/prognostic value. |
