circRNA-disease information
Expression pattern:
UP
Associated gene: JAK2, STAT3, p-STAT3, FUS, CTCF
Associated microRNA: miR-485-3p
Biological function: promotes proliferation, migration and invasion; enhances tumour growth and metastasis
Molecular mechanism: Acts as a ceRNA to sponge miR-485-3p and upregulate JAK2, increasing p-STAT3; directly interacts with and recruits FUS to facilitate nuclear translocation of p-STAT3; transcriptionally enhanced by CTCF
Biological pathway or process:

JAK/STAT (promotes); proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); ceRNA regulation (promotes)

Detected method:
Q
S
Validation methods:

RT-qPCR; RNA-seq; FISH / smFISH; Nuclear-Cytoplasmic Fractionation; RNase R Treatment; divergent primers PCR; Sanger Sequencing; Actinomycin D / DRB Stability Assay; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); RNA Pull-Down; Transfection; CCK8; Colony Formation Assay; Transwell Assay; Wound Healing Assay; Western Blot; Co-IP; ChIP / ChIP-seq; IHC (Immunohistochemistry); IF (Immunofluorescence); H&E Staining; In Vivo Animal Model; Clinical Sample Validation; Survival Analysis; ROC Analysis; Bioinformatics Analysis

Clinical significance:

High expression of circSPARC was associated with advanced TNM stage, lymph node metastases, and poor survival; circSPARC might serve as a potential diagnostic and prognostic biomarker

Description:

circSPARC (hsa_circ_0004104), derived from SPARC, is upregulated in CRC tissues and plasma and promotes CRC proliferation, migration, invasion, tumor growth and metastasis. It acts as a cytoplasmic ceRNA sponging miR-485-3p to increase JAK2 expression and enhance JAK2/STAT3 signaling (increased p-STAT3), and also recruits the RBP FUS to facilitate nuclear translocation of p-STAT3. High circSPARC is associated with advanced clinicopathologic features and poorer overall survival, with potential diagnostic/prognostic value.

Confidence score:

0.9033
