circRNA-disease information
Expression pattern:
UP
Associated gene: CDK16, JAK2, STAT3
Associated microRNA: miR-324-5p
Biological function: Promotes HCC malignant behaviors (cell growth/proliferation, cell cycle progression, migration, invasion) and inhibits apoptosis; GL63 exerts anti-tumor effects partly by downregulating circZNF83.
Molecular mechanism: Acts as a ceRNA sponge for miR-324-5p to derepress CDK16, thereby affecting JAK2/STAT3 signaling; GL63 downregulates circZNF83 to inhibit this axis.
Biological pathway or process:

JAK/STAT (inhibits); proliferation (promotes); cell cycle (promotes); apoptosis (inhibits); migration (promotes); invasion (promotes); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RT-qPCR; Clinical Sample Validation; Bioinformatics Analysis; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Transfection; CCK8; Colony Formation Assay; Cell Cycle Assay; Annexin V/PI Flow Cytometry; Western Blot; Transwell Assay; In Vivo Animal Model

Clinical significance:

Description:

circZNF83 is up-regulated in HCC tissues and cell lines and functions as an oncogenic circRNA. It sponges miR-324-5p to increase CDK16 expression and support malignant phenotypes, and GL63 suppresses HCC partly by downregulating circZNF83 to inhibit the miR-324-5p/CDK16 axis and inactivate JAK2/STAT3 signaling.

Confidence score:

0.7052

Other information
Title:

A curcumin analog GL63 inhibits the malignant behaviors of hepatocellular carcinoma by inactivating the JAK2/STAT3 signaling pathway via the circular RNA zinc finger protein 83/microRNA-324-5p/cyclin-dependent kinase 16 axis.
