circRNA-disease information
Expression pattern:
UP
Associated gene: PD-L1, Ago2
Associated microRNA: miR-377-3p
Biological function: promotes tumorigenesis, proliferation, colony formation, migration and invasion in NSCLC
Molecular mechanism: ceRNA mechanism: circ_0000284 sponges miR-377-3p to up-regulate PD-L1
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); ceRNA regulation (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Nuclear-Cytoplasmic Fractionation; Clinical Sample Validation; Transfection; CCK8; Colony Formation Assay; Transwell Assay; Western Blot; In Vivo Animal Model; Luciferase Reporter Assay; RIP (RNA Immunoprecipitation); Survival Analysis

Clinical significance:

High circ_0000284 expression is associated with low survival and may indicate NSCLC occurrence/progression

Description:

circ_0000284 (circHIPK3) is up-regulated in NSCLC tissues/cell lines, mainly localized in the cytoplasm, and promotes NSCLC growth and metastatic behaviors. Mechanistically, it functions as a ceRNA to sponge miR-377-3p, thereby relieving repression of PD-L1 and enhancing PD-L1 expression; high circ_0000284 correlates with poorer patient survival.

Confidence score:

0.8323

Other information
Title:

Circular RNA circ_0000284 plays an oncogenic role in the progression of non-small cell lung cancer through the miR-377-3p-mediated PD-L1 promotion.
