In A549 lung cancer and HCT116 colorectal cancer cells, circPVT1 promotes oncogenic phenotypes (proliferation, migration, apoptosis resistance and EMT). Mechanistically, circPVT1 antagonizes let-7 to relieve repression of NRAS via the NRAS 3′UTR, and blocking let-7/NRAS binding rescues NRAS reduction and several phenotypes caused by circPVT1 knockdown.
