circRNA-disease information
Expression pattern:
UP
Associated gene: STAT3, HK2
Associated microRNA: miR-515-5p
Biological function: Promotes GC cell proliferation and enhances the Warburg effect (increased glucose consumption, lactate production, ATP quantity, and ECAR; reduced OCR); knockdown represses tumor growth in vivo.
Molecular mechanism: circCUL3 acts as a ceRNA (miRNA sponge) for miR-515-5p to upregulate STAT3; STAT3 transcriptionally activates HK2 by binding the HK2 promoter, promoting aerobic glycolysis (Warburg effect).
Biological pathway or process:

glycolysis (promotes); proliferation (promotes); other pathway/process (promotes); ceRNA regulation (promotes)

Detected method:
Q
M
Validation methods:

Microarray; RT-qPCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; Sanger Sequencing; Nuclear-Cytoplasmic Fractionation; FISH / smFISH; Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Western Blot; ChIP / ChIP-seq; In Vivo Animal Model; Survival Analysis; Bioinformatics Analysis; Clinical Sample Validation; Cohort Study

Clinical significance:

High circCUL3 expression correlates with advanced clinical stage and unfavorable prognosis/overall survival in GC patients.

Description:

circCUL3 is up-regulated in gastric cancer tissues and cell lines and is associated with worse overall survival. It promotes GC cell proliferation and aerobic glycolysis (Warburg effect) by sponging miR-515-5p to increase STAT3, which then transcriptionally upregulates HK2; circCUL3 knockdown suppresses xenograft tumor growth.

Confidence score:

0.8784
