circRNA-disease information
Expression pattern:
UP
Associated gene: KAT5, c-MYC, histone H3 lysine 27 acetylation (H3K27ac), RNA polymerase II
Associated microRNA: -
Biological function: promotes NSCLC cell proliferation and viability; inhibits apoptosis; promotes cell cycle progression; promotes tumor growth in vivo
Molecular mechanism: circRHOT1 directly interacts with KAT5 and recruits it to the c-MYC promoter to enhance H3K27ac and RNA polymerase II enrichment, thereby epigenetically increasing c-MYC transcription/expression
Biological pathway or process:

proliferation (promotes); apoptosis (inhibits); cell cycle (promotes); other pathway/process (promotes)

Detected method:
Q
Validation methods:

RT-qPCR; Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; FISH / smFISH; Transfection; MTT; Colony Formation Assay; EdU Staining; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Western Blot; ChIP / ChIP-seq; RNA Pull-Down; RIP (RNA Immunoprecipitation); In Vivo Animal Model; H&E Staining; IHC (Immunohistochemistry); TUNEL

Clinical significance:

-

Description:

circRHOT1 is up-regulated in NSCLC tumor tissues and lung cancer cell lines and promotes proliferation and tumor growth while suppressing apoptosis. Mechanistically, circRHOT1 binds KAT5 and facilitates its recruitment to the c-MYC promoter, increasing transcription-associated H3K27ac/RNA polymerase II enrichment and elevating c-MYC expression.

Confidence score:

0.8151

Other information
Title:
