In lymph node-negative CRC with synchronous liver metastasis (LNLM1), hsa_circ_0124554 is up-regulated and promotes invasion/migration and in vivo metastasis. Mechanistically, it directly binds AKT, inhibits AKT ubiquitination, sustains AKT phosphorylation, and activates AKT-associated downstream signaling (e.g., GSK-3beta and mTOR). Clinically, higher expression is associated with vascular invasion/liver metastasis and poorer prognosis, and plasma levels may help distinguish LNLM1 from LNLM0.
