circRNA-disease information
Expression pattern:
UP
Associated gene: IRS2
Associated microRNA: miR-613
Biological function: circFAM126A promotes NSCLC cell proliferation, migration, invasion, cell cycle progression and tumor growth, and inhibits apoptosis; knockdown of circFAM126A suppresses these malignant phenotypes and induces apoptosis and cell cycle arrest.
Molecular mechanism: circFAM126A functions as a miRNA sponge for miR-613 to upregulate IRS2 expression, thereby promoting NSCLC progression.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); cell cycle (promotes); EMT (promotes); ceRNA regulation (promotes)

Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Microarray; Clinical Sample Validation; RNA Pull-Down; Luciferase Reporter Assay; Transfection; MTT; EdU Staining; Colony Formation Assay; Annexin V/PI Flow Cytometry; Cell Cycle Assay; Transwell Assay; Wound Healing Assay; In Vivo Animal Model; IHC (Immunohistochemistry); Western Blot; Survival Analysis; Bioinformatics Analysis

Clinical significance:

High circFAM126A expression is associated with worse overall survival in NSCLC patients and may be helpful for NSCLC diagnosis.

Description:

circFAM126A is up-regulated in NSCLC tissues and cells and high expression is associated with poorer survival. Functionally, circFAM126A acts as an oncogenic circRNA that promotes proliferation, migration, invasion, cell cycle progression, EMT-related marker changes and xenograft tumor growth while suppressing apoptosis. Mechanistically, it sponges miR-613 to upregulate IRS2, forming a circFAM126A/miR-613/IRS2 regulatory axis.

0.8712
