Expression pattern:
DN
Associated gene: -
Associated microRNA: miR-7, miR-1290, miR-671
Biological function: Not directly involved in mediating IMiD resistance; epigenetically silenced in resistant MM cells
Molecular mechanism: Epigenetic silencing associated with promoter CpG island hypermethylation of LINC00632 T3 promoter; partial reactivation by DNMT and EZH2 inhibition
Biological pathway or process:

drug resistance (other); ceRNA regulation (other)

Detected method:
S
Validation methods:

RNA-seq; Bioinformatics Analysis; RT-qPCR; Transfection; CCK8; Annexin V/PI Flow Cytometry

Clinical significance:

-

Description:

ciRS-7 (CDR1as) is markedly down-regulated in acquired lenalidomide- and pomalidomide-resistant multiple myeloma cell models. Its loss correlates with hypermethylation of the LINC00632 T3 promoter CpG island and can be partially reversed by DNMT and EZH2 inhibition, but ciRS-7 knockdown alone does not change IMiD sensitivity.

Confidence score:

0.4852
