circRNA-disease information
Expression pattern:
UP
Associated gene: Rap1b, VEGFR2, PI3K, AKT, VEGF-A, CD31
Associated microRNA: miR-30c-5p
Biological function: Promotes LC progression, proliferation, migration, invasion, tumor growth and angiogenesis; inhibits apoptosis; exosomal circPVT1 enhances endothelial cell migration and tube formation.
Molecular mechanism: Exosomal circPVT1 acts as a miR-30c-5p sponge to upregulate Rap1b, activating VEGFR2 and PI3K/AKT signaling to promote angiogenesis.
Biological pathway or process:

VEGF/VEGFR (promotes); PI3K/AKT (promotes); angiogenesis (promotes); proliferation (promotes); migration (promotes); invasion (promotes); apoptosis (inhibits); ceRNA regulation (other); drug resistance (other)

Detected method:
Q
Validation methods:

Back-Splice Junction PCR / divergent primers PCR; RNase R Treatment; Sanger Sequencing; RT-qPCR; FISH / smFISH; Clinical Sample Validation; IHC (Immunohistochemistry); Transfection; CCK8; Colony Formation Assay; Wound Healing Assay; Transwell Assay; Tube Formation Assay; In Vivo Animal Model; Western Blot; Luciferase Reporter Assay; ELISA; Bioinformatics Analysis; Survival Analysis; Cohort Study

Clinical significance:

High circPVT1 expression is correlated with advanced T stage and lower survival rate; exosomal circPVT1 may serve as a valuable biomarker, prognostic marker and therapeutic target for LC.

Description:

circPVT1 is upregulated in laryngeal cancer tissues, cell lines and patient plasma/serum exosomes, and high expression is associated with disease progression and poorer survival. LC cell-derived exosomal circPVT1 is transferred to endothelial cells, where it sponges miR-30c-5p to increase Rap1b and activate VEGFR2/PI3K/AKT signaling, promoting angiogenesis. Targeting circPVT1 suppresses tumor growth and angiogenesis and may improve the therapeutic effect of cetuximab.
