circRNA-disease information
Expression pattern:
UP
Associated gene: METTL3, YTHDF1, FOXM1, PCNA, p21
Associated microRNA: miR-761, miR-4778-3p
Biological function: circRPS6KC1 promotes proliferation and cell cycle progression, inhibits cellular senescence, and supports tumor growth in prostate cancer; suppression of circRPS6KC1 induces G1/G0 arrest, decreases DNA replication, increases SA-beta-gal-positive senescent cells, and reduces xenograft tumor growth.
Molecular mechanism: METTL3 and YTHDF1 mediate m6A modification and stabilization of circRPS6KC1. Cytoplasmic circRPS6KC1 functions as a miR-761 sponge to regulate FOXM1 expression; FOXM1 transcriptionally regulates PCNA and influences p21 degradation in a p53-independent manner.

Description:

circRPS6KC1 is an up-regulated human circRNA in prostate cancer derived from RPS6KC1. METTL3/YTHDF1-mediated m6A modification stabilizes circRPS6KC1, and circRPS6KC1 promotes proliferation and suppresses cellular senescence by sponging miR-761 to regulate FOXM1. FOXM1 then regulates PCNA transcription and p21 protein degradation in a p53-independent pathway.
