circRNA-disease information
Expression pattern:
UP
Associated gene: TEAD1, DERL1, Ago2
Associated microRNA: miR-873-5p
Biological function: circTTBK2 acts as an oncogenic circRNA in NSCLC, promoting cell proliferation, migration, invasion and tumor growth, and its overexpression is associated with TNM stage, lymph node metastasis and poorer survival.
Molecular mechanism: circTTBK2 functions as a cytoplasmic miRNA sponge for miR-873-5p in a RISC manner, relieving miR-873-5p-mediated repression of TEAD1; TEAD1 transcriptionally activates DERL1 to promote NSCLC malignancy.
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); ceRNA regulation (promotes); Hippo/YAP (other)

Detected method:
Q
Validation methods:

RNase R Treatment; Actinomycin D / DRB Stability Assay; RT-qPCR; Clinical Sample Validation; Transfection; CCK8; EdU Staining; Wound Healing Assay; Transwell Assay; In Vivo Animal Model; IHC (Immunohistochemistry); RIP (RNA Immunoprecipitation); ChIP / ChIP-seq; Luciferase Reporter Assay; Western Blot; Cohort Study; Survival Analysis; Bioinformatics Analysis

Clinical significance:

Overexpressed circTTBK2 in NSCLC tissues indicates poor prognosis; circTTBK2 expression is positively associated with TNM stage and lymph node metastasis, implying a potential therapeutic strategy for NSCLC treatment.

Description:

circTTBK2 is a human TTBK2-derived circRNA that is up-regulated in NSCLC tissues and cell lines, and high expression is associated with poorer prognosis, TNM stage and lymph node metastasis. Functionally, circTTBK2 promotes NSCLC cell proliferation, migration, invasion and xenograft tumor growth. Mechanistically, cytoplasmic circTTBK2 sponges miR-873-5p to increase TEAD1, which transcriptionally activates DERL1, forming the circTTBK2/miR-873-5p/TEAD1/DERL1 oncogenic axis.
