| Biological function: |
Promotes HCC cell proliferation, migration and invasion; reduces apoptosis; enhances tumorigenicity and metastasis in vivo. |
| Molecular mechanism: |
Acts as a nuclear scaffold forming a ternary complex with EZH2 and STAT3, promoting EZH2-induced STAT3 methylation and phosphorylation to activate STAT3 signaling; activated STAT3 binds the circ-LRIG3 promoter to increase circ-LRIG3 transcription, creating a positive feedback loop. |

| Clinical significance: |
High circ-LRIG3 is associated with aggressive clinicopathological features, is an independent risk prognostic factor for overall survival, and plasma circ-LRIG3 is a sensitive and specific non-invasive diagnostic indicator for HCC. |
| Description: |
circ-LRIG3 (hsa_circ_0027345) is upregulated in HCC tissues/cells/plasma and functions as an oncogenic nuclear circRNA. It scaffolds EZH2 and STAT3 to enhance STAT3 methylation/phosphorylation and activate STAT3 signaling, and activated STAT3 increases circ-LRIG3 transcription to form a positive feedback loop, promoting proliferation, invasion and metastasis and inhibiting apoptosis. Clinically, high circ-LRIG3 indicates poor prognosis and plasma circ-LRIG3 shows diagnostic potential for HCC. |
