circRNA-disease information
Expression pattern:
UP
Associated gene: S100A14, Ago2, PTK2
Associated microRNA: miR-139-3p
Biological function: promotes cervical cancer cell proliferation, migration, invasion, angiogenesis, EMT-related malignant behaviors, and tumor growth in vivo
Molecular mechanism: circ_0003221 acts as a cytoplasmic miRNA sponge for miR-139-3p and positively regulates S100A14 expression through the miR-139-3p/S100A14 regulatory axis
Biological pathway or process:

proliferation (promotes); migration (promotes); invasion (promotes); metastasis (promotes); angiogenesis (promotes); EMT (promotes); ceRNA regulation (other)

Detected method:
Q
Validation methods:

RNase R Treatment; RT-qPCR; Clinical Sample Validation; RIP (RNA Immunoprecipitation); Luciferase Reporter Assay; Transfection; CCK8; EdU Staining; Transwell Assay; Tube Formation Assay; Western Blot; Nuclear-Cytoplasmic Fractionation; In Vivo Animal Model; Survival Analysis; Bioinformatics Analysis

Clinical significance:

high circ_0003221 expression was related to poor 5-year overall survival and circ_0003221 may be a possible diagnostic biomarker for cervical cancer

Description:

circ_0003221 is up-regulated in cervical cancer tissues and cell lines and high expression is associated with poor 5-year overall survival. Functionally, circ_0003221 promotes cervical cancer cell proliferation, migration, invasion, angiogenesis, EMT-related malignant phenotypes, and xenograft tumor growth. Mechanistically, cytoplasmic circ_0003221 sponges miR-139-3p and thereby positively regulates S100A14 expression through the circ_0003221/miR-139-3p/S100A14 axis.
